چکیده
The first application of thioacid capture ligation in protein synthesis is described. Two histone H3 proteins were synthesized in which a 30 min ligation reaction gave the protein products in good yields.

Facile synthesis of site-specifically acetylated and methylated histone proteins: reagents for evaluation of the histone code hypothesis.
The functional capacity of genetically encoded histone proteins can be powerfully expanded by posttranslational modification. A growing body of biochemical and genetic evidence clearly links the unique combinatorial patterning of side chain acetylation, methylation, and phosphorylation mainly within the highly conserved N termini of histones H2A, H2B, H3, and H4 with the regulation of gene expr...

A common post-translational modification (PTM) of proteins is lysine acetylation. This is an especially ubiquitous PTM in the histones of chromatin, and is important for helping to regulate both structural and mechanistic aspects of chromatin. The fundamental unit of chromatin is called the nucleosome and is made up of DNA that wraps around a histone protein octamer. Protruding from the nucleos...
متن کاملProtein synthesis by native chemical ligation: expanded scope by using straightforward methodology.
The total chemical synthesis of proteins has great potential for increasing our understanding of the molecular basis of protein function. The introduction of native chemical ligation techniques to join unprotected peptides next to a cysteine residue has greatly facilitated the synthesis of proteins of moderate size. Here, we describe a straightforward methodology that has enabled us to rapidly ...

Posttranslational modification (PTM) of histones plays a central role in genome regulation. Engineering histones with defined PTMs on one residue or on multiple residues is crucial for understanding their function within nucleosomes and chromatin. We introduce a sequential native chemical ligation strategy that is suitable for the preparation of fully synthetic histone proteins, allowing for si...

Two in one : Total chemical synthesis by the connection of three fragments via two consecutive ligation reactions carried out in a one-pot manner provided histone protein H2A homogeneously phosphorylated at Tyr57 (see scheme). The assembly of phosphorylated H2A and ubiquitinated H2B with histone proteins H3 and H4 into a nucleosome provided direct evidence for the inhibition of the activity of ...
